That depends on if your tuner is a chromatic tuner or not. A chromatic tuner will show you the exact note that is played when a string is plucked -- and show you flat or sharp. If all you have is an EADGBE tuner it will be more difficult. Most ukulele are tuned GCEA. (Some older players prefer ADF#B) The "G" string can be either low (Low G tuning) or high (Re-entrant or standard tuning) depending on which types of strings you had strung on your ukulele -- (hint) low-G strings are thicker than all the others.

Chromatic tuners are relatively inexpensive nowadays and some can be found for under $25.
